Pharmacokinetics

Absorption: Polythiazide has an oral bioavailability is 100%, pre-systemic (first-pass) metabolism accounts for 0.5% (±0.5%).

Distribution: Polythiazide has a volume of distribution is 4.1l/kg. plasma protein binding is 85 %.

Elimination: it is metabolised via Hepatic, plasma half-life is 25 hr, renal excretion is 25 %.

Indications

Polythiazide is primarily indicated in conditions like Diabetes insipidus, Edema, Heart failure, Hypercalciuria, Hypertension, Oedema.

Contraindications

Polythiazide is contraindicated in conditions like Diabetes mellitus, Hypokalaemia.

Side Effects

Polythiazide in overdosage may give rise to further complications which include Atherosclerosis .

Polythiazide produces potentially life-threatening effects which include Hyponatremia, Hypokalemia. Careful monitoring is needed with proper medical management.

The signs and symptoms that are produced after acute overdosage of Polythiazide are Hypotension, Volume depletion, Renal failure, Hypokalemia , Hyponatremia.

Polythiazide is known to cause more or less tolerable side effects which are usually transient. These include Dizziness, Gout, Headache, Impotence, Nausea.

Warnings / Precautions

Thiazide diuretics increase sensitivity to sunlight, avoid prolonged sun exposure, use a sunscreen and wear protective clothing. It should be used during pregnancy or lactation only if clearly needed. It should be used with caution in patients with medical history especially if have gout or diabetes, or any allergy especially to sulfa medications. Avoid any drug that increase heart rate or cause excitation like decongestants (found in cough and cold products) because it may counter-act blood pressure medicine.
