Estradiol is a naturally occuring sex hormone (oestrogen). estradiol is used along with progesterone for contraception, menstrual disorder, and for the suppression of certain cancers and also for the prevention of osteoporosis.

Absorption: Estradiol. has an oral bioavailability is 60.5% (±9.5%), pre-systemic (first-pass) metabolism accounts for 83% (±7%).

Distribution: Estradiol. has a volume of distribution is 9-5 liters. plasma protein binding is 97 - 99 %.

Elimination: it is metabolised via Gut wall and Hepatic, plasma half-life is 20-70 min, renal excretion is 80 %.

Side Effects

Estradiol. in overdosage may give rise to further complications which include Irritability , Gynacomastia , Uterine bleeding , Tachyphylaxis.

Estradiol. produces potentially life-threatening effects which include Endometrial carcinoma, Breast cancer. Careful monitoring is needed with proper medical management.

The signs and symptoms that are produced after acute overdosage of Estradiol. are Vomiting, Dizziness, Headache, Speech problem, Breast Lump, Redness in one leg, Chest crushing pain, Shortness of breath, Double Vision, Coughing up blood.

Estradiol. is known to cause more or less tolerable side effects which are usually transient. These include Breast tenderness, Depression, Fluid retention, Headache, Nausea and vomiting, Weight gain.

Warnings / Precautions

High doses of estrogen used in treatment of malignant disease should be used cautiously in patients with cerebrovascular disorder, coronary artery disease or venous thromboembolism. Estrogens should be used with caution in children, because premature closure of epiphysis may occur resulting in inhibited linear growth and small stature.

Interference in Pathology

Estradiol. may interfere in the diagnosis of Thyroid Function Test , Plasma Cortisol Test , Urinary 17-Oxogenic Steroid Test .
