Lisuride

Lisuride is a dopamine receptor agonist. Lisuride is semisynthetic ergot derivative belonging to the 8a-aminoergoline group. Lisuride is effective in the treatment of parkinsons disease especially in the latter stages of disease when the treatment fails.It is reported to have serotonergic activity and has been used in disorders associated with hyperprolactinaemia. Lisuride is not available in any combination preprations. Lisuride is administered orally.

Absorption: Lisuride has an oral bioavailability is 50% (±50%), pre-systemic (first-pass) metabolism accounts for 85% (±5%).
Distribution: Lisuride has a volume of distribution is 2.3-2.4 l/kg. plasma protein binding is 70%.
Elimination: plasma half-life is ~8 hr.

Indications
Lisuride is primarily indicated in conditions like Cushing's syndrome, Hyperprolactinaemia, Migraine prophylaxis, Neuroleptic malignant syndrome, Parkinsonism, Parkinson's disease.
Lisuride is also indicated for secondary conditions like Acromegaly, Galactorrhoea.
Contraindications
Lisuride is contraindicated in conditions like Peripheral vascular disease, Pituitary disease, Coronary heart disease.

Side Effects
Lisuride in overdosage may give rise to further complications which include Delirium , Vivid dreams, Confusion , Memory disturbance , Visual hallucination , Drowsiness , Frequent & vivid dreams .
The signs and symptoms that are produced after acute overdosage of Lisuride are Exanthemia, Satyriasis, Hallucinations .
Lisuride is known to cause more or less tolerable side effects which are usually transient. These include Abdominal pain, Dizziness, Itching, Nausea, Vomiting , Weight gain.
