Domperidone is a dopamine blocking agent. Domperidone is antiemetic agent. Domperidone is also used to treat nausea and vomiting induced by cancer chemotherapy.

Absorption: Domperidone has an oral bioavailability is 69.75% (±23.25%), pre-systemic (first-pass) metabolism accounts for 43.5% (±41.5%).
Distribution: Domperidone has a volume of distribution is 5.7 l/kg. plasma protein binding is 91-93%.
Elimination: it is metabolised via extensively by liver & Gut, plasma half-life is 12-16 hr (oral), 7.5 hr (parentral), renal excretion is major 31% and 66% in feces.

Indications
Domperidone is primarily indicated in conditions like Acute nausea and vomiting, Delayed gastric emptying, Dyspepsia, Functional dyspepsia, Gastric distension pain due to pressure on the stomach, Gastro-oesophageal reflux disease, Nausea and vomiting, Nausea and vomiting (chemotherapy induced), Nausea and vomiting (L-dopa induced), Non-ulcer dyspepsia, Reflux oesophagitis.
Domperidone is also indicated for secondary conditions like Cytotoxic chemotherapy, Heart burn, Lactation.
Contraindications
Domperidone is contraindicated in conditions like Allergy, Hepatic impairment, Serious resistant GI mycoses.

Side Effects
Domperidone in overdosage may give rise to further complications which include Parkinsonism , Extrapyramidal dystonic reactions , Tardive dyskinesia .
Domperidone produces potentially life-threatening effects which include Seizures , Dysrhythmias. Careful monitoring is needed with proper medical management.
The signs and symptoms that are produced after acute overdosage of Domperidone are Convulsions , Coma, GI disturbances, Extrapyramidal effects .
