Chlorpropamide is oral hypoglycemic agent. Chlorpropamide is used in the treatment of non-insulin dependent diabetes mellitus. Chlorpropamide is used in conjunction with proper diet and exercise to decrease blood sugar levels.

Absorption: Chlorpropamide has an oral bioavailability is 100%, pre-systemic (first-pass) metabolism accounts for 9%.

Distribution: Chlorpropamide has a volume of distribution is 0.14-0.21 l/kg. plasma protein binding is 57-96%.

Elimination: it is metabolised via extensive by Liver 80%, plasma half-life is 24-48hr, renal excretion is major.

Indications

Chlorpropamide is primarily indicated in conditions like Diabetes insipidus, Diabetes mellitus, Diabetes mellitus type-ii.

Chlorpropamide is also indicated for secondary conditions like Softening and dilatation of cervix before mechanical cervical dilatation.

Side Effects

Chlorpropamide in overdosage may give rise to further complications which include Thrombocytopenia , Cholestatic jaundice , Aplastic anemia , Hemolytic anemia , Cholestatic jaundice , Aplastic anemia , Hemolytic anemia , Fictitious hypoglycemia, Agranulocytosis , Exfoliative dermatitis , Eosinophilic pulmonary infiltrates , Hemolytic anemia, Aplastic anemia.

Chlorpropamide produces potentially life-threatening effects which include Hypoglycemia , Hypoglycemia, Hypoglycemia. Careful monitoring is needed with proper medical management.

The signs and symptoms that are produced after acute overdosage of Chlorpropamide are Coma, Seizures, Hypothermia, Hypoglycemia.

Chlorpropamide is known to cause more or less tolerable side effects which are usually transient. These include Abdominal pain, Diarrhea, Dyspepsia , Facial flushing, Headache, Hyponatremia.

Warnings / Precautions

Chlorpropamide should be used with caution in patients with medical history, especially if have high blood pressure or liver or kidney disease. Avoid alcohol while taking this medication. Photosensitization may occur, caution patients to take appropriate measure (e.g.wear protective clothing and a sunscreen) against exposure to UV or sunlight. It should be used in pregnancy or lactation only if clearly needed. Insulin therapy may be necessary during pregnancy.
