Milrinone (Lactate) is a phosphodiesterase III inhibitor similar to amrinone, with +ve ionotropic and vasodilator activity. It potentiate effects of cyclic adenosine monophosphate(cAMP). Milrinone (Lactate) is administered intravenosly in the management of heart failure(severe) unresponsive to other form & in acute heart failure after cardiac surgery.

Absorption: Milrinone (Lactate) has an oral bioavailability is 82.5% (±2.5%).

Distribution: Milrinone (Lactate) has a volume of distribution is 0.32 l/kg. plasma protein binding is 70 %.

Elimination: it is metabolised via hepatic(12%), plasma half-life is 2-3 hr, renal excretion is 83 % unchanged.

Side Effects

Milrinone (Lactate) in overdosage may give rise to further complications which include Hypotension , Bronchospasm , Hypotension , Bronchospasm , Angina , Anaphylaxis , Supraventricular arrhythmia, Ventricular arrhythmia.

Milrinone (Lactate) produces potentially life-threatening effects which include Arrhythmias, Hypotension. Careful monitoring is needed with proper medical management.

The signs and symptoms that are produced after acute overdosage of Milrinone (Lactate) are Hypotension.

Milrinone (Lactate) is known to cause more or less tolerable side effects which are usually transient. These include Angina, Headache, Hypokalemia , Hypotension , Nausea, Palpitation, Rashes , Somnolence , Thrombocytopenia, Tremor.

Warnings / Precautions

Blood pressure, heart rate, ECG, fluid and electrolyte balance should be monitored during therapy. It should be used with caution in patients with severe obstructive aortic or pulmonary valvular disease or with hypertrophic cardiomyopathy and in children and in patients with decreased renal function,hypokalemia,hypotension.

Interference in Pathology

Milrinone (Lactate) may interfere in the diagnosis of Liver Function Test (LFT) .
