Vigabatrin is anticonvulsant agent. Vigabatrin is analogue of GABA (gamma amino-butyric acid). GABA is an inhibitory neurotransmitter, which is thought to be deficient in epilepsy, and able to stabilise discharges from epileptic foci. Vigabatrin is used in treatment of epilepsy, especially when not controlled by other drugs.

Absorption: Vigabatrin has an oral bioavailability is 85% (±5%), pre-systemic (first-pass) metabolism accounts for 0.5% (±0.5%).

Distribution: Vigabatrin has a volume of distribution is 0.85 l/kg. plasma protein binding is negligible.

Elimination: plasma half-life is 7-8 hr.

Indications

Vigabatrin is primarily indicated in conditions like Epilepsy, Infantile spasms.

Vigabatrin is also indicated for secondary conditions like Iron deficiency anaemia.

Contraindications

Vigabatrin is contraindicated in conditions like Ventricular arrhythmias resistant to other treatment.

Side Effects

The signs and symptoms that are produced after acute overdosage of Vigabatrin are Coma, Vertigo , Unconsciousness, Tremor.

Vigabatrin is known to cause more or less tolerable side effects which are usually transient. These include Ataxia, Confusion, Depression, Diplopia, Dizziness, Fatigue, Headache, Irritability, Nervousness, Psychosis, SomnolenceX.

Warnings / Precautions

Vigabatrin should be used with caution during pregnancy. It should be used with caution in elderly and patient with renal (kidney) impairment. Care is required when withdrawing vigabatrin therapy. Caution is warranted in patients with history of psychosis or behavioral problems.
