Pharmacokinetics

Absorption: Orciprenaline (Sulphate) has an oral bioavailability is 20% (±20%), pre-systemic (first-pass) metabolism accounts for 37.5% (±37.5%).

Distribution: Orciprenaline (Sulphate) has a volume of distribution is 700 litre.

Elimination: it is metabolised via GI Wall, plasma half-life is 6 hr, 20 hr, renal excretion is 40 %.

Indications

Orciprenaline (Sulphate) is primarily indicated in conditions like Asthma, Bronchitis, Obstructive airway disease.

Orciprenaline (Sulphate) is also indicated for secondary conditions like Bradycardia, Reversible airways obstruction.

Contraindications

Orciprenaline (Sulphate) is contraindicated in conditions like Cardiac arrhythmia, Diabetes mellitus, Myocardial infarction, Thyrotoxicosis, Ischaemic heart disease, Cystic fibrosis, Aortic stenosis, Quadriplegia.

Side Effects

Orciprenaline (Sulphate) in overdosage may give rise to further complications which include Intolerable muscle tremors , Extra systoles .

Orciprenaline (Sulphate) produces potentially life-threatening effects which include Ventricular arrhythmias, Deterioration of asthma. Careful monitoring is needed with proper medical management.

The signs and symptoms that are produced after acute overdosage of Orciprenaline (Sulphate) are Nausea, Tachycardia , Headache, Tremor, Restlessness , Anxiety , BP changes, Excitement.

Orciprenaline (Sulphate) is known to cause more or less tolerable side effects which are usually transient. These include Dizziness, Hyperglycemia, Hypokalemia , Hypotension , Nausea, Nervousness, Palpitation, SweatingX, Tachycardia, Taste disturbance, Tremor.

Warnings / Precautions

Orciprenaline should not be used in the patient with hypothyroidism, diabetes mellitus, hypertension, cardiac arrhythmias, ischemic heart disease, seizures or hypersensitivity to sympathomometics (pheochromocytoma). The safe use of drug during pregnancy has not been established. The inhaled Orciprenaline should be used with great caution in the children under the age of 12 years.
