Amodiaquine is a 4-aminoquinoline antimalarial with an action and uses similar to that of chloroquine. Amodiaquine is not recommended for the prophylaxis of malaria. Amodiaquine is used to treat malaria (intermittent or remittent disease characterized by attacks of chills, fever, and sweating and caused by a parasitic protozoan transferred to the human bloodstream by an anopheles mosquito).

Absorption: Amodiaquine has an oral bioavailability is 82.5% (±2.5%), pre-systemic (first-pass) metabolism accounts for 82.5% (±2.5%).

Distribution: Amodiaquine has a plasma protein binding is extensive.

Elimination: it is metabolised via via liver, plasma half-life is 5.2 hr.

Indications

Amodiaquine is primarily indicated in conditions like Malaria, Nephrogenic and partial pituitary diabetes insipidus.

Contraindications

Amodiaquine is contraindicated in conditions like Malaria prophylaxis.

Side Effects

Amodiaquine in overdosage may give rise to further complications which include Liver damage .

Amodiaquine produces potentially life-threatening effects which include Blood dyscrasias, Lupus erythematosus. Careful monitoring is needed with proper medical management.

Amodiaquine is known to cause more or less tolerable side effects which are usually transient. These include Abdominal cramps, Diarrhea, General malaise, Irregular heart beat, Nail-beds pigmentation, Nausea, Palate pigmentation, Skin pigmentation, Termors, Tinnitus, Vomiting , Weakness.

Warnings / Precautions

It should be used with caution in patients allergic to peanuts, not to be used on broken, infectious skin. It should be used with caution in blockage of the gut (intestinal obstruction).
