Hydroxyprogesterone is intermediate formed in the conversion of cholesterol to cortisol, androgen and estrogens, used in contraception (to prevent pregnancy). Hydroxyprogesterone is also used to treat dysmenorrhea, irregular uterine bleeding and in the management of endometriosis.

Absorption: Hydroxyprogesterone has an oral bioavailability is 90%, pre-systemic (first-pass) metabolism accounts for 50% (±5%).

Elimination: it is metabolised via liver, plasma half-life is 4 hr.

Indications

Hydroxyprogesterone is primarily indicated in conditions like Abortion prevention, Habitual abortion, Preterm labour.

Hydroxyprogesterone is also indicated for secondary conditions like Endometrial cancer.

Contraindications

Hydroxyprogesterone is contraindicated in conditions like Breast cancer, Vaginal bleeding, Hyperlipidemia, Liver carcinoma.

Side Effects

Hydroxyprogesterone in overdosage may give rise to further complications which include Allergic reaction chest pain pepression.

Hydroxyprogesterone is known to cause more or less tolerable side effects which are usually transient. These include Diarrhea, Edema, Flatulence, GI upset, Local pain, Pre-existing acne & urticaria, Weight gain.

Warnings / Precautions

Hydroxyprogesterone should not be used during pregnancy or lactation. It should be used with caution in patients with any pre-existing liver disease, diabetes, seizure disorder, kidney disease, heart trouble, asthma or a history of depression, migraines, blood clots, miscarriage or breast cancer or any allergy. Because this medication may increase sensitivity to sunlight, avoid prolonged sun expose, wear a sunscreen and protective clothing when outdoors. Diabetic patients should monitor urine and blood sugar closely while taking this medication as it can reduce glucose tolerance.

Interference in Pathology

Hydroxyprogesterone may interfere in the diagnosis of Measurement of Progesterone in Plasma .
