Absorption: Sulphamethizole has an oral bioavailability is 85% (±15%).

Distribution: Sulphamethizole has a plasma protein binding is 90%.

Elimination: it is metabolised via Hepatic, plasma half-life is 1.5-3 hours, renal excretion is 60% in 5 hours and 90% in 10 hours.

Indications

Sulphamethizole is primarily indicated in conditions like Urinary tract infection.

Sulphamethizole is also indicated for secondary conditions like Otitis media, Prophylaxis of rheumatic fever.

Contraindications

Sulphamethizole is contraindicated in conditions like Renal failure, Hepatic failure.

Side Effects

Sulphamethizole in overdosage may give rise to further complications which include Dermatitis , Epidermal necrolysis , Stevens johnson syndrome , Anuria.

Sulphamethizole produces potentially life-threatening effects which include Anemia. Careful monitoring is needed with proper medical management.

The signs and symptoms that are produced after acute overdosage of Sulphamethizole are Fever, Rashes , Pruritus , Exfoliative dermatitis , Nephritis.

Sulphamethizole is known to cause more or less tolerable side effects which are usually transient. These include Anorexia , Diarrhea, Fever, Hypersensitivity , Nausea and vomiting.

Warnings / Precautions

In patients using it,adequate fluid intake is necessary to reduce risk of crystalluria. Sulphamethizole should be discontinued immediately if rash appears. Sulphamethizole should be given with care to patients with severe hepatic or renal failure,asthma,allergy. Sulphamethizole is bacteriostatic, a structural analogue of PABA that interferes with nucleic acid synthesis.

Interference in Pathology

Sulphamethizole may interfere in the diagnosis of Urine tests for Catecholamines, dopa, glucose, ketones, hippuric acid , Creatinine determination by Alkaline Picrate Method .
